Guitar into External Signal Processor "SIGNAL IN". Then use the main SIGNAL OUT (very top right of patch panel) to go out to your amp, mixer, etc. It's important to set all the knobs as you see here; especially for the filter cutoffs and VCO mixer levels. Once you get this working, use the filter cutoffs to shape the tone (there are actually two pairs of filters, one within the signal processor, and one within the main synth architecture.) Also, if you turn up the VCO mixer volumes, you'll hear the MS-20's oscillators droning; you can adjust their pitch using the tuning knobs, or by playing notes on the keyboard.
